Q: I’m a size UK 12. Will medium be correct choice?

Q: Would it be suitable to wear over the backpack too?

Difficult to say as I do not know how big your rucksack is but with the adjustments you can make with it around the waist, providing you bought one sufficiently larger than you, I can't see why not. Place the backpack on, slip the LED Vest over your head and position it to fit then do up the side adjust strap around the waist. It may not be perfect but I'd take a guess to say it would do the job well enough. I'd maybe say if you are already a well built/sized man, having a rucksack on too may take too much of the slack in the shoulder blade/over head area which is not adjustable but is capacious. I bought an XL to fit over my clothing and leather motorcycle jacket. I'm well built though only 5'8. It fits well over me and the clothing and is actually still big around the chest/mid sect so could accommodate more, I have the waist tightened up to the smallest adjustment (it does mean there is a small amount of waist strap material flapping).
