Does it work if i put it on over nail polish

This works best when put on just before you go to bed .I would putting on over a coat of polish would not penetrate the nail or nourish it.

My cuticles are very dry. Does it make sense to apply multiple times a day?

Well, sure, if you don't mind your fingers being a bit oily several times a day. Even after you rub it in, it takes awhile to fully soak in. I do mine at night.
