Yes. Put bees in jar with powered sugar. Shake with screen on jar onto white paper or cardboard. Count mites and determine if need treated.

Yes, the bee's, (200 to 300) depending on the size sample used, will be killed if using 99% alcohol as recommend. I suggest reading about it and other options.
