I have 275/40/20, will they fit well?

I have them on a set of 245/50/20 so they should fit.

Will this for a 305/35/22?

Might be a bit too small. I have them on 20’s but 22’s May be too big.

Is it possible to buy a single one of these anywhere?

Thank you for asking about this, Jeffery! These covers are only available in a set of four; we do apologize for any inconvenience this causes.

Can it fit my 255/35/-21inch tires?

Manufacturer says covers should fit tires 22" to 31" in diameter. Your tires are 28" in diameter (search for tire size calculator online). Thus they ought to fit.
