Will it accommodate 256gb SSD taken out from MacBook pro 13.3 inch 2015

No. This will not work. All old MAC between 2013 to 2015 - uses 12 + 16 pin and it will not fit into these enclosures.. There is a convertor for those (old apple 12+16 to sata converter) and that can be used.

Does it supports gen4 nvme ssd

All most all pcie gen 4 device is backward compatibil , so a pcie gen 4 device connected to pcie gen 3 slot then it will work at gen 3 speed , this enclosure data transfer speed depends on your uab port generation
