No, this is a splitter not an insert cable, it divides a single Stereo (or TRS balanced) into 2 Stereo/TRS signals. You need something like the Hosa STP 203.

Q: Can I connect two headphones?

Yes. You can use tow pair of headphones. Expect some gain loss, so may not be as loud as with just one pair of headphones.

Q: Will this work with the inputs on guitar cables and amps?

Yes, it is standard 1/4 phono guitar size

Q: Can I connect two microphones into one input on my amplifier to go out to 1 to 4 headphones?

It should work. You will have two mono signals, one for each mic. You should hear one mic on the left channel and one mic on the right channel. I used the TISNO for splitting a stereo signal headphone output from the mixer to two different headphones.
