There's nothing in the blood to make it react. The test looks for acid phosphatase which is in seminal fluid in high concentrations, while other body fluids, like female secretions have very low concentrations.

What if someone uses ammonia to destroy the semen?

If its been destroyed there is nothing to test. But how do you know it is destroyed? Do a test on a semen sample that you clean away with ammonia. This will tell you if it is test-able. If it is positive there will be a bright purple color that appears quickly.
