Can this be used on the shimano sh-am501 mtb shoes?

I am not familiar with the Shimano SH-AM 501 MTB shoe to be able to answer your question.

Will this work with the MT5 shoes. I am looking for replacement part for that shoe for covering the hole where the cleats would go

Having seen the MTS, and owning similar Shimano shoes with the SPS conversion, I can say confidently that these will work. Keep in mind that there may be a little variance and not an identical fit, small gaps may be present. If a little large, one can file the rubber to improve the fit. They worked for me.

Yes, they should work on any SPD shoe. They work on Pearl Izumi shoes for sure. They could be trimmed if too big for cleat recess.
